SALK Population Information

The SALK Population at NASC are single, segregating flank-tagged T3-generation T-DNA lines. The population was generated by Dr. Joseph Ecker, at the The Salk Institute in California, USA.
Plasmid Construct
The construct that has been introduced into Arabidopsis ecotype Columbia (Col) via Agrobacterium tumefaciens vacuum infiltration is the pROK2 vector, a derivative of the pBIN19 vector, as shown in Figure 1 (below).

Selectable Marker
As seen in Figure 1, the selectable marker is the NPTII (kanamycin resistance) gene. However, after several generation of growth, some of the lines show silencing of this gene. Thus, it is not unusual for a mutant line to not express the drug resistance phenotype.
